For a "Custom Calculated Symbol", you would have to create the symbol in the main instance and then have it "flow down" to the sub-instance. This is because the sub-instances get their data from the main instance, so the main instance would need to know of that symbol. This should not be an issue, as long as you have the following option set to "Yes": General Settings Window: Use Symbol Settings in New Instances when Started (Global Settings >> General Settings >> General >> New Instances) You do also need to restart the second instance after you create the symbol in the main instance. -- As to speed/effectiveness to executing trades on the sub-instance, there will be a very small (probably on the order of a few microseconds) difference since the sub-instance has to send the order through the main instance. But in terms of the total time to send an order to the exchange, this will be negligible.
