There is no reason to change the Tick Size or the Price Display Format, as both the E-Mini S&P 500 (ES) and the Micro E-Mini S&P 500 (MES) have the same settings (.25 Tick Size, therefore a Price Display format of 0.01). The Currency Value per Tick does need to be changed to the value of the symbol you have set in the "Trade and Current Quote Symbol", so entering $6.25 is correct for this item.
