regarding a second instance of sierra, if the "Always Use Data Folder Within Main Folder" is deselected and the Data Files Folder location is changed and saved, when the second instance is restarted these settings revert back to the default.

This is not a bug. This is by design. You cannot use any other folder for a sub instance.

I cannot accommodate a 50 GB data file in the SierraChart folder with the current configuration of my computer. Can I just move the folder to a different drive and change the "Target" and "Start in" fields in the shortcut?

You can move the main instance of Sierra Chart to a different drive. But the sub instances have to be located with it as well.
