Today I noticed that after every restart of SC the Global Profit and Loss Management is disabled! That is a quiet substantial feature I rely on and for years GPNL was active right from the start.

I updated to 2337 but still the same issue causing all GPNL functions to be disabled.
- Flatten when Profit rules met
- Trail daily profit
- Net loss management
- Flatten after set time

Is there any new setting I'm not aware of or how can this be fixed?

There is a setting on the Global Profit and Loss Management in the bottom right-hand corner that states Disable Global Profit/Loss Management On Startup. You would want this to be disabled in order to have the functionality running when you start Sierra Chart. Refer to the following:
Global Profit/Loss Management: Disable Global Profit/Loss Management on Startup

Same here. Disable GPNL is "disabled" so GPNL should be enabled on startup.
